It's likely black beard algae (BBA) on the java fern.
Harlequin rasbora do spawn upside down under the leaves of plants, so there's a possibility that an egg may be attached to the BBA. However, this would be unlikely and be unusual behaviour for them even if there were an egg there.
Some fish have preferred locations in certain areas of the tank. My elderly last-remaining harlequin has a preferred spot above the centre of a clump of java fern - and woe betide anyone (particularly any female of another species) who enters that territory.... she shoos them away.
