No, it will give a false positive (as will almost every test on the market). This is because the tests unlock the "safe" ammonium and convert back to toxic ammonia, so if there is "locked" non-toxic ammonium in the water the result will be positive for ammonia for some time until the filter has processed it.
You can overcome this by using a Seachem Ammonia Alert in your tank. This will only respond to toxic ammonia and gives a true view of the actual ammonia level in your water. |

If you tried an ammonia alert in your tank right now hopefully it would read 0ppm. If the tank has been cycling for 3 weeks hopefully the ammonia will soon start to drop, but often fish in cycles can take longer due to the frequent water changes.

You will eventually get to 0. However, there's another factor to consider. If your water supplier uses chloramine then when you use Prime (or any dechlorinator which also treats chloramine) the chemical bond between the chlorine and ammonia that make up that compound is broken. The chlorine is then neutralised, and in the case of Prime the ammonia is "locked" into a safe form (ammonium). That can result in a false positive on tests depending on tank volume/water change volume and amount of ammonia released by detoxification of chloramine so it's beneficial to use an ammonia alert to be certain. |
